The Monster Girls (まものガールズ) are a series of 5 characters introduced in Puyo Puyo!! Quest. They are all based on and share names with mythological creatures. The 5 consider each other close friends and often play and spend time together.

Lilim is a Devilling girl described as having cow horns and bat wings. She is selfish and entitled, and often gets upset if others don't comply to her frequent demands. However, if someone proves to be reliable to her, she can become quite the handful. While overbearing, her earnest and serious side emerges whenever it's called for. She believes that the willingness of others to listen to her is a result of her attractiveness, and thinks no one else on Pwurp Island can compare to her beauty. She feels challenged when others are called cute, and feels the need to assert herself as the most beautiful. Her demanding personality is in contrast with her own willingness to fulfill the requests of others, which is said to make her popular among some, although she will usually demand rewards for completing tasks. Due to her strong dislike of sunlight, she's mostly active during the night. However, on days where the sun is harsh, her selfish tendencies are said to subside. On these days, she also becomes demotivated, but as long as she isn't under the sun, she's always giving it her all. She often refers to herself as "Lady Lilim" (リリンさま) and requests that others do the same. She claims to have good night vision. She doesn't like feeling left out.

Lamia is a girl with the lower body of a snake. She has a spirited and enthusiastic personality. She's a glutton and is constantly eating anything she can, but she especially likes sweets, including candy, cakes and sundaes. Her gluttony and love for sweets tends to be all she ever thinks about, and her peppy attitude can take a sudden shift if she hasn't eaten properly. She's also prone to getting angry if someone interrupts her eating. She's willing to do just about anything if she can get her hands on sweets as a reward. She is said to be skilled at moving around stealthily, but the sound of her grumbling stomach tends to blow her cover.
She is generally believed to be a separate entity from the Lamia that appears as an enemy in the Madou Monogatari series due to their differing designs and personalities. However, the Puyo Puyo 25th Anniversary Book features Lamia alongside other characters first appearing in main series titles, which may imply the two characters are one in the same, as is the case with Succubus.

Empusa is a female monster with the appearance of a praying mantis. She has a passionate, combative personality and a rather rough manner of speech. She's a hard worker, and is constantly challenging others as well as seeking opportunities to become stronger. Her belief in hard work leads her to take things seriously, and she places high value in a winning mentality. However, her hardworking characteristics can lead her to go overboard and believe her efforts aren't good enough, a fact she herself is aware of. Her serious attitude and fighting spirit creates a high level of expectation in her opponents, insisting that they shouldn't show weakness in front of her. She also gets excited if she identifies an opponent with a strong spirit. She hates to lose and believes in payback if she does. She puts a lot of care into her dual sickles and makes sure they're always shining without a scratch on them.

- Empusa is based off of and named after the empusa mantis.
- As the other Monster Girls have origins in Greek mythology, Empusa's name could also be referencing the Greek mythological creature Empusa, although the two appear to share no relation.
- Lamia's Active Skill name, Buchikamasu (ブチカマース), is derived from the Japanese phrase buchikamasu, meaning to slam.
- In Japanese, kama also means sickle, which is the weapon Empusa wields.
- She is an excellent cook and specializes in an odd dish called a "flourling", which can be seen in [★6] Exspatulary Empusa's art.
- She wears a happi instead of an apron when she cooks because they bolster her spirit, which she believes is more important.
- She is shown to sometimes serve street food during festivals on Pwurp Island.
- Empusa is associated with the Victory Headband, a headband which is said to strengthen power during the most crucial moments of battles.

Sphinx is a monster girl with the lower body of a lion and bird wings on her arms. Her temperament is subdued and lazy, and she's prone to falling asleep very easily if she isn't careful. This isn't a bother to her, though, sleeping and lazing around are her hobbies, and she can be impossible to move out of bed if she doesn't have the motivation. She's not often driven to perform tasks, and any of her work must be preceded by a nap. If she wishes, Sphinx is also capable of sleeping for incredibly long periods of time, and she can also doze off at any time, including in the heat of battle. Overly complicated subjects bore her. Despite appearing to own several pillows, she will sometimes use her wings as pillows for napping. She has a perpetual bedhead and dreams frequently in her sleep. She tends to yawn a lot.

- Sphinx is based off of the mythological Sphinx, which is also her namesake.
- While the Sphinx originates from Egyptian mythology, Puyo Puyo's Sphinx seems to be based more heavily on Greek tradition, since the female qualities as well as the eagle wings both originate from the Greek version of the creature.
- Sphinx also wears an ankh, the Egyptian symbol of life, in her hair.
- Sphinx's Active Skill name, Makhadda (マハッダ), is derived from the Arabic word maḵadda, meaning pillow.
- Sphinx's Fluffy Pillow is said to be so comfortable that it's hard to live without it once it's slept on.
- Sphinx shares a name with a card from Arle no Bouken: Mahou no Jewel. There is no relation between the two characters.

Gorgon is a monster girl with snakes for hair. She is an earnest and hardworking person. She is also caring and worries about others. Anyone who looks into her eyes will be turned into stone, so she shields her eyes for the sake of others. As a child, she would wear a visor to keep her eyes covered, but now she simply keeps her eyes closed herself. She's able to see through the eyes of the snakes on her heads, though each snake has its own sentience and will sometimes fight, so it can be hard for her to keep them coordinated. This leads her to be somewhat clumsy, as she trips easily and is prone to getting lost. The eyesight she gains from them is poor, so she relies on her snakes to give her directions. She's able to somewhat understand what her snakes are saying to her. Each snake has a name, though the only known names currently of her six hair snakes are Hebinosuke (ヘビ之助) and Hebisaburo (ヘビ三郎), which are implied to be the two to her bottom right (the sad snake and the neutral one underneath it). She seems to have a desire to show her eyes to others.

- Gorgon is based off of the mythological species Gorgon of which she is named after.
- Gorgon shares a name with Gorgon, a cow-like enemy from the MSX version of Madou Monogatari 2 who doesn't at all resemble the mythological Gorgon.
- Gorgon's Active Skill name is Petra (ペトラ), which is Latin for stone.[2]
- She is associated with the Maiden Gaze, a strange bracelet which allows one to sense oncoming danger.
- The two known names of Gorgon's hair snakes start with hebi, the Japanese word for snake. This could imply that all of Gorgon's hair snakes follow this naming convention.

- Empusa is said to admire Lilim for her guts and assertiveness. She often challenges her to competitions, which shows she views Lilim as a worthy opponent.
- Empusa and Sphinx
- Empusa remains friends with Sphinx despite their contrasting personalities. Empusa will challenge her to Puyo Battles, indicating her respect for Sphinx as an opponent. Sphinx states that she'll miss Empusa when she announces her intent to go on a journey.
